Forodesine/呋咯地辛 生物活性

描述 Forodesine (BCX-1777) is identified as a potent, orally active inhibitor of purine nucleoside phosphorylase (PNP), showing IC50 values ranging from 0.48 to 1.57 nM across human, mouse, rat, monkey, and dog PNP. It notably hinders human lymphocyte proliferation and induces apoptosis in leukemic cells by elevating dGTP levels[1].[2].
体内研究

Forodesine exhibits excellent oral bioavailability (63%) in mice and, at a single 10 mg/kg dose, raises dGuo levels to approximately 5 μM[2].

In the hu-PBL-SCID mouse model, Forodesine notably extends lifespan by two-fold or more[2].
体外研究

Treatment with Forodesine at 10-30 μM for 24 and 48 hours partially inhibits the proliferation of RPMI-8226, MOLT-4, and 5T33MM cells. While it does not impact MM cells at 24 hours, it decreases the viability of MOLT-4 cells by 40%[1].

In the presence of 2'-deoxyguanosine (dGuo, 3-10 μM), Forodesine significantly inhibits the proliferation of human lymphocytes activated by IL-2, mixed lymphocyte reaction (MLR), and phytohemagglutinin (PHA), with IC50 values below 0.1-0.38 μM[2].
