NP-313 生物活性

描述

NP-313 is a powerful antithrombotic compound that effectively suppresses platelet aggregation and activation. It achieves its effects through dual inhibition of thromboxane A2 synthesis and selective suppression of SOCC-mediated Ca2⁺ inward flow^[1].

体内研究

NP-313 (i.v., 4-16 µg/g) notably extends occlusion time (TTO), increases bleeding time, and suppresses platelet aggregation induced by collagen (10 µg/mL) in male ICR mice ^[1].

体外研究

NP-313 (0-80 μM) demonstrates dose-dependent inhibition of platelet aggregation. The highest inhibition levels observed for platelet aggregation induced by thrombin and A23187 were approximately 80% and 60%, respectively ^[1].

NP-313 demonstrated concentration-dependent inhibition (0-8 μM) of P-selectin expression and thromboxane B2 (TXB2) production in human platelets stimulated by collagen or thrombin. Furthermore, NP-313 inhibited COX-1 and TXA2 synthase with IC50 values of 1.5 and 3.9 µM, respectively ^[1].
