Bleomycin Sulfate/硫酸博来霉素 生物活性

描述 Bleomycin Sulfate is a glycopeptide antitumor antibiotic with antineoplastic efficacy[1]. Bleomycin sulfate treatment (3.5-4.0 mg/kg; intra-tracheal) on day 0, body weights decrease by day 4 then increases by Day 7 through the end of the study. Intra-tracheal bleomycin produced a statistically significant increase in lung hydroxyproline levels as well as increase in right caudal lung lobe mass[2]. Bleomycin sulfate (intratracheally; 2.5 mg/kg; 1.25 mg/ml, approximately 50 µl per mouse) induces pulmonary fibrosis in male C57BL/6 mice (8 weeks old, average weight approximately 24.5 g) [3].
体外研究 Bleomycin sulfate (BLM) has been selected as the most well-studied micronucleus (MN) inducer in human lymphocytes with different mechanisms of genotoxicity. The most common types of Bleomycin-induced DNA damage are single and double strand breaks and single depyrimidine/dehydropyrimidine sites. At the same time Bleomycin is a true radiomimetic compound, almost exactly resembling the genetic effects of ionising radiation [4]. The IC50 value of Bleomycin sulfate against the UT-SCC-19A cell line was 4.0±1.3 nM. Both UT-SCC-12A and UT-SCC-12B were more resistant to Bleomycin sulfate (BLM); IC50 values were 14.2±2.8 nM and 13.0±1.1 nM, respectively [5]. Bleomycin sulfate (50, 100 μM; 24, 48 h) induced pulmonary fibrosis in RLE-6TN cells (50 μM) and A549 cells (100 μM) [6].
