Spectinomycin (Dihydrochloride Pentahydrate) is a broad-spectrum aminocyclitol antibiotic that is highly effective in the treatment of uncomplicated gonorrhea. It is a good alternative drug in the treatment of penicillin-allergic patients[3]. Aminomethyl spectinomycins are active against Gram-negative and Gram-positive bacterial pathogens. Aminomethyl spectinomycins are promising new candidates for development of therapeutics against biodefense bacterial agents[4]. Spectinomycin selectively inhibits protein synthesis in cells and in extracts of Escherichia coli. However, spectinomycin is not bactericidal and causes no detectable misreading of polyribonucleotides[5]. The kinetic analysis demonstrated that spectinomycin acts as a mixed noncompetitive inhibitor for the td (T4 thymidylate synthase gene) intron RNA with a K(i) of 7.2 mM. Increasing the spectinomycin concentration raised the K(m) values with the corresponding decrease of V(max) and k(cat) values[6].
