D-Luciferin potassium salt is the water-soluble potassium salt form of D-Luciferin. D-Luciferin is the substrate for the enzyme Firefly Luciferase. It can produce bioluminescence through oxidation by the enzyme luciferase. The typical emission spectrum for luciferase is in the yellow-green region (550–570 nm), with a peak at 562 nm at basic media (pH~7.5–7.8)[1]. For D-Luciferin potassium salt possesses water solubility, it is usually in in vivo bioluminescence imaging[2].
1. Prepare a fresh stock solution of Luciferin at 15mg/mL in DPBS. Filter sterilize through a 0.2 µm filter.
2. Inject 10 µL/g of body weight. Each mouse should receive 150 mg Luciferin/kg body weight. (e.g. For a 10 g mouse, inject 100 µL to deliver 1.5 mg of Luciferin.)
3. Inject the Luciferin intra-peritoneally (i.p.) 10-15 minutes before imaging.
Note: A Luciferin kinetic study should be performed for each animal model to determine peak signal time.

In vitro bioluminescence imaging assay procedure:[3]:
1. Prepare a 100mM (100-200X) D-Luciferin reserve solution in sterile water. Stir well. Use immediately, or one-time packaging, store at -20℃, avoid freeze-thaw cycle, avoid light.
2. The working solution of 0.5-1mM D-Luciferin was prepared in preheated medium.
3. Remove the medium from the cultured cells.
4. Luciferin working solution was added to the cells and incubated at 37 °C for 5-10 minutes before imaging.
