AY 9944 生物活性

描述 AY 9944 is a specific cholesterol biosynthesis inhibitor that inhibits 7-dehydrocholesterol Δ7 reductase (DHCR7) with an IC50 of 13 nM. AY 9944 causes hypocholesterolaemia and accumulation of 7DHC. At high doses, AY 9944 also inhibits the sterol Δ7-Δ8 isomerase in cultured embryos, leading to the accumulation of cholesterol-8-en-3β-ol[1][2][3].AY 9944 affects the type of free sterols in keratinocytes at a concentration of 1 µg/ml for 15 h. AY 9944 inhibits the synthesis of cholesterol and desmosterol, but increases the synthesis of 7-dehydrocholesterol and oleanol[2].

AY 9944 动物研究

Animal study At a dose of 7.5 mg/kg, administered every 6 days from postnatal day 2 to postnatal day 20, i.h., AY 9944 reduced brain cholesterol in Long-Evans hooded rats until 400 days after cessation of treatment. Brain cholesterol was reduced and 7-dehydrocholesterol was elevated in all brain regions of the rats, and AY 9944 had more severe effects on changes in plasma and liver steroids in female rats[3].
