SB756050 was well tolerated, readily absorbed, and exhibited nonlinear pharmacokinetics, with smaller than dose-proportional increases in plasma exposure above 100 mg and no significant change in exposure when combined with sitagliptin.The pharmacodynamic effects of SB756050 were highly variable within and between dose groups, with blood glucose increasing at the two lowest doses and not decreasing at the two highest doses. Despite the different plasma profiles of the gut hormones, the glucose-lowering effect of SB756050 sitagliptin was comparable to that of sitagliptin alone^[1].
