描述 TAPI-1 is a TACE (ADAM-17) and MMP inhibitor. It could inhibit the processing of TNF. Treatment with TAPI-1 at concentration ranging in 12.5-200μM led to a dose-dependent reduction of TNF stimulated by LPS in both THP-1 cells and peripheral blood T cells. Administration of 1mg TAPI-1 subcutaneously could protect the mice from the lethal dose of LPS. TAPI-1 at concentration of 2μM could suppress sIL-6R production in THP-1 macrophages and THP-1 cells after 12-hour treatment. And also this treatment significantly suppressed IL-1β-induced increases in the expression OPN mRNA and production of OPN, but exhibited no effect on OPN without IL-1β stimulation.
