alpha-Asarone is a naturally occuring inhibitor of both HMG-CoA reductase (IC50 = 3 mM) and certain isoforms of the cytochrome P450 superfamily of enzymes (IC50 = 55.2 and 65.2 μg/ml for CYP2D6 and CYP3A4, respectively), with neuroprotective, anti-oxidative, anticonvulsive and cognitive enhancing action.
Alpha-Asarone is a major bioactive component isolated from Acorus gramineus, an herbal medicine that possesses anti-oxidant, anti-hyperlipidemic and anti-inflammatory effects. Alpha-Asarone exhibited acetylcholinesterase (AChE) inhibitory effect in a dose dependent manner. The values of 18.5%, 28.9%, and 66.0% were obtained for tested concentrations of 0.25, 0.5, and 1 mg/ml, respectively.in vivo, hyperlocomotion could be alleviated by administration (10 mg/kg for 15 days) of alpha-Asarone in fragile X mental retardation gene (Fmr1) knockout (KO) mice, which had observable disturbances in the striatal cholinergic system[3].
