PJ34 生物活性

描述

PJ34 inhibits PARP enzyme activity with an IC50 of 110±1.9 nM. For comparison of neuroprotective properties with other PARP inhibitors in PC12 cells, PJ34 is assessed using an LDH assay. Additionally, PJ34 treatment significantly and concentration-dependently reduces cell death at concentrations ranging from 10-7 to 10-5 M^[1].

PJ34 动物研究

Animal study

To assess its potency and efficacy relative to other PARP inhibitors, PJ34 is tested at doses of 3.2 and 10 mg/kg, respectively. At a dose of 3.2 mg/kg, PJ34 significantly reduces cortical damage by 33%; however, the higher dose of 10 mg/kg demonstrates a reversed effect, with only a 17% reduction^[1].PJ34 (25 mg/kg) reduces TNF-α mRNA levels in ischemic animals by 70%, bringing them on par with those in sham or naive animals. In ischemic mice treated with PJ34, E-selectin mRNA levels decrease by 81% and ICAM-1 mRNA levels by 54% compared to vehicle-treated ischemic mice^[2].
