Chicago Sky Blue 6B is a potent inhibitor of L-glutamate uptake into synaptic vesicles and also inhibits macrophage migration inhibitory factor (MIF) with IC50 of 0.81 μM.

Direct Blue 1, known as Chicago Sky Blue 6B, is employed to quench background autofluorescence in immunofluorescence histochemistry. It acts as a potent and competitive inhibitor of VGLUT and can block the binding of Aβ to small molecule PrP ligands. Direct Blue 1 exhibits anti-inflammatory properties^[1]^[2]^[3]^[4]^[5]^[6].
