Deoxynivalenol/脱氧雪腐镰刀菌烯醇 生物活性

描述 Deoxynivalenol is a trichothecene mycotoxin that frequently contaminates cereal grains, leading to immune dysregulation and growth impairment in animals. Incubation of U937 monocytes with 500 ng/mL deoxynivalenol induced the phosphorylation of p38 at 15min and 30min post-treatment. Deoxynivalenol treatment also induced the sequential association of p38 with 40S and 60S ribosomal subunits in U937 monocytes. In RAW 264.7 cells, 250 ng/mL deoxynivalenol induced p38 association with the pooled RS + M fractions, evidenced by an increased level of total p38 in this fraction at 5–30 min post-treatment. Deoxynivalenol at 250 ng/mL also promoted the phosphorylation of JNK and ERK in RAW 264.7 cells from 15–30 min[3]. In mice, the consumption of 0.5 ppm deoxynivalenol (= 0.075 mg/kg/day) for 8 weeks reduced food intake and 2 ppm purified deoxynivalenol (= 0.3 mg/kg/day) resulted in decreased growth in comparison to non-deoxynivalenol group[4].
作用机制 DON induces ribotoxic stress response in mononuclear phagocytes by binging to ribosome, facilitating the interaction of p38 with the ribosome, inducing p38 phosphorylation, and eventually affecting downstream events, including mRNA transcription, protein translation and the activation of apoptotic effectors.
