描述 | Toll-like receptors (TLR) are a family of type I transmembrane receptors essential for the innate immune response. SM-324405 is a TLR7 agonist with an EC50 of 50.1nM in a reporter gene assay using HEK293 cells expressing human TLR7. The half-life (T1/2) of SM-324405 in human plasma was 2.6min. SM-324405 induced type I interferon via TLR7 activation in human peripheral blood mononuclear cells with a minimum effective concentration of 10nM. In Brown Norway rats sensitized by ovalbumin, intratracheal administration with SM-324405 at 0.1 or 1mg/kg inhibited eosinophil influx into bronchoalveolar lavage fluid in a dose-dependent manner as compared to the control group, while systemic IFN induction was not observed in SM-324405-treated rats[1]. |
