描述 | 5-TAMRA (5-Carboxytetramethylrhodamine), SE and its conjugates yield bright, pH-insensitive orange-red fluorescence (approximate excitation/emission maxima ~546/579) with good photostability. 5-TAMRA is a purified monomer. It is widely used as a marker of peptides and proteins. In some complex biological studies, the reproducibility of experiments is more critical than material consumption, and it has become the preferred fluorescent labeling reagent. There is only slight difference between 5-TAMRA and 6-tamra in structure, but it will significantly affect the biological properties of the conjugate formed. TEMPO-5-TAMRA fluorescence quenching is a promising method to measure short distance changes within single biomolecules. In combination with distance calculations using molecular mechanics modeling, the experimental results from steady-state ensemble fluorescence and fluorescence correlation spectroscopy measurements both show an exponential decrease in the quenching rate constant with the dye-quencher distance in the 10-30 A range[3]. |
