描述 | Dihydroorotate dehydrogenase (DHODH) is the rate-limiting enzyme in the uridine monophosphate (UMP) biosynthetic pathway and mostly located in the inner membrane of mitochondria, catalyzing the transformation of dihydroorotate to orotate dependent of Mg2+ and ATP[1]. DHODH-IN-11 (Compound 14B), a derivative of Leflunomide, is a weak inhibitor of DHODH with a pKa of 5.03. The weak potency of DHODH-IN-11 may be due to the adverse stereochemistry of the oxime structure[2]. |
