Interleukin-1 receptor-associated kinase 4 (IRAK4) is a kinase required for innate immune responses induced by Toll-like receptors and the interleukin-1 receptor family members. IRAK4-IN-4 (compound 15 in patent CN107163044A) is an IRAK4 inhibitor with an IC50 value of 2.8nM. Cyclic GMP-AMP synthase (cGAS) is a cytosolic protein that acts as an intracellular innate immune sensor that detects double-stranded DNA. IRAK4-IN-4 also exhibits inhibitory activity towards cGAS with an IC50 value of 2.1nM[1].
