描述 | Type II NADH: quinone oxidoreductase (NDH-2) is a membrane-bound bi-substrate enzyme that catalyses the cytoplasmic oxidation of NADH and reduction of quinone in the membrane. HQNO is a potent mitochondrial NDH-2 inhibitor in many species. Wide type NDH-2 had an IC50 value of 10.5 ± 1.3 μM HQNO in the presence of 400 μM menadione (MD). In the presence of 50 μM MD, the IC50 value for HQNO decreased slightly to 7.3 ± 1.2 μM and near complete inhibition was observed with > 50 μM HQNO[1]. | ||
作用机制 | HQNO is nested into the slot-shaped tunnel of the Q-site, in which the quinone-head group is clamped by Q317 and I379 residues, and hydrogen-bonds to FAD[1]. |
