Retinol-binding protein 4 (RBP4) is a serum protein that transports retinol from the liver to other tissues. A1120 is a high affinity, non-retinoid ligand for RBP4 with a Ki value of 8.3nM. The IC50 values of A1120 for human RBP4 and mouse RBP4 are 90nM and 66nM, respectively. In B6D2F1 mice, a hybrid F1 strain between C57Bl/6 and DBA/2, single oral administration of 30mg/kg A1120 decreased serum levels of RBP4 and retinol with the peak reduction observed at 12 hours after treatment. In B6D2F1 mice fed a high-fat diet for 6 weeks, administration of A1120 (as a diet admixture at 30mg/kg) for additional 4 – 6 weeks did not affect glucose levels following the oral glucose challenge[1].
